reactivity. As anti-mitochondria1 antibodies can bind complement we used a complement-fixation test. Results from preliminary tests were difficult to interpret. The Paracoccus membranes had strong anti-complementary activity, which had to be diluted out. Also the sera available from patients with primary biliary cirrhosis were themselves anti-complementary up to quite high dilutions. Nevertheless, the sera tested did appear to demonstrate complement-fixing reactivity against both mitochondria1 membranes and Paracoccus membranes when compared with the corresponding normal serum controls. Further tests will be necessary to confirm the specificity of the reaction between Puracoccus denitrificans and sera from subjects with primary biliary cirrhosis. Corresponding tests are also clearly desirable with membranes from other aerobes and from anaerobes such as Streptococcus faecalis. These studies should also be extended to sera of mice immunized with syngeneic mitochondria (Davey et al., 1975) and to mice immunized with bacterial membrane preparations. Confirmation of a cross-reaction between anti-mitochondria1 antibody and bacterial membranes could throw new light on the nature of the mitochondrial antigen, and might also have implications as to alternative origins of anti-mitochondria1 activity in sera in human subjects.

The conjugation of arylacetic acids in the pigeon compared with the hen.

In the domestic hen phenylacetic acid is excreted conjugated mainly with ornithine (Totani, 1910), but in the pigeon it is excreted conjugated with taurine and glycine (James et al., 1972). This difference between the hen and pigeon has now been further investigated with several arylacetic acids. The compounds used were 14C-carboxy-labelled phenylacetic acid, 4-chloroand 4-nitrophenylacetic aci...

The Conjugation of a - Naphthylacetic Acid in vivo

Aromatic and arylalkyl carboxylic acids commonly undergo metabolic conjugation prior to excretion, the conjugating agent being either a carbohydrate or an amino acid. Among mammalian species the carbohydrate is usually glucuronic acid, but in the case of the amino acids, one or more of glycine, glutamine or taurine may be used, and the particular pattern of amino acid conjugation depends upon t...

Bile acid conjugation and hepatic taurine concentration in rats fed on pectin.

A relationship between bile acid conjugation and hepatic taurine concentration was investigated in rats fed on citrus pectin. When rats were fed on the diets containing varying amounts of pectin (10, 30, 60 and 100 g/kg dietary levels), biliary excretion of bile acids increased as the dietary levels of pectin increased. The increase was entirely due to the glycine-conjugated bile acids. The bil...

Duodenal bile acid conjugation patterns and dietary sulphur amino acids in the newborn.

Patterns of glycine and taurine conjugation of bile acids in duodenal juice have been studied in 20 low birth weight babies at 12 and 22 days of age. They were fed from birth on one of three feeds containing different amounts of taurine and its precursors, cysteine and methionone. Taurine conjugation predominated in those fed on human milk throughout the first three weeks of life. In those fed ...

The biochemical basis for the conjugation of bile acids with either glycine or taurine.

All animals, except for the placental mammals, conjugate their bile acids exclusively with taurine. However, in certain of the placental mammals, glycine conjugates are also found. The basis for the appearance of glycine conjugation among the placental mammals was investigated. The reaction of choloyl-CoA with glycine and taurine, as catalysed by the soluble fraction from guinea-pig liver, had ...
